Single-molecule manipulation provides a highly sensitive way to study micromechanics and interactions of bio-molecules at a single-molecule level. In the first part of this talk, I will introduce the basic principles of single-molecule manipulation methods. In the second part of the talk, I will introduce their applications in studies of micromechanics of DNA, protein, and their interactions. In particular, I will focus on our recent finding on the properties of DNA-binding by H-NS-like proteins which are important for global gene silencing and chromosomal DNA organization in bacterial. We found these proteins can form rigid nucleoprotein filaments that are important for gene silencing. As the gene-silencing H-NS nucleoprotein filaments do not form on curved DNA, this finding also imply that anti-silencing of H-NS silenced genes can be achieved by introducing large DNA curvature by other proteins. In addition, I will also introduce how a novel DNA structure is uncovered by single-molecule manipulation experiments.
